ADAM8 or CD156 is a disintegrin metalloproteinase. Recent works have demonstrated that ADAM8 can be detected in urines from breast cancer patients and in sputum from individuals with inflammatory lung conditions. ADAM8 also plays a role in inflammatory lung conditions such as COPD and asthma. In addition, ADAM8 expression in breast cancer was correlated with tumor dissemination and metastasis.

We were able to show that processing of our ADAM8 substrates, Pepdab013 (Flsub13) and Pepdab005(Flsub005), by urines from patient breast cancer patients with invasive and metastatic disease, was significantly higher than normal controls. When the two substrates were used together, the sensitivity and specificity of the assay was greater than 90% for each.

In addition to selling ADAM8 substrates that are sensitive and specific, Biozyme, Inc. now sells human recombinant ADAM8 expressed and purified from HEK293 cells. The recombinant ADAM8 is supplied in 10 and 20ug aliquots as a lyophilized powder.  A 10ug aliquot is enough for approximately 50 assays using PEPDAB013, a ADAM8 specific substrate, or PEPDAB005, a general ADAM substrate that is more sensitive than PEPDAB013.

The Recombinant ADAM8 that is carrier free (CF),  has greater than 90% purity for the catalytic mature form. The recombinant ADAM8 preparation also contains approximately 10% of the pro-form of the enzyme. Unlike other preparations for commercially available recombinant ADAM8, our enzyme does not require a pre-activation step.
