MMP Substrates

BioZyme Inc. has many mmp substrates to choose from. We have selective MMP13 substrates, MMP9 substrates, MMP1 substrates, and MMP2 substrates. In addition we have a general mmp substrate, PEPDAB008,  that is useful for assays with MMP1, MMP2, MMP3, MMP7, MMP8, MMP10, MMP12, MMP13, MMP14, and MMP16. Each substrate we have varies in its selectivity pattern for individual MMP family members, making it possible to mathematically determine how much activity is due to individual proteinases. The program that does this was invented by individuals at MIT and is termed proteolytic activity matrix analysis or PrAMA. However, BioZyme Inc also sells selective substrates for some of the MMP family members, that can be used in biological samples and cell based assays where PrAMA is not necessary.

The mmp substrates can also be used as simple tools for screening purposes, inhibition assays, and determination of active enzyme concentrations and most of them have been used in cell based assays to assess activity due to MMP2, MMP9, MT1-MMP or MMP14 and MMPs in general.
All of our MMP FRET substrates have Dabcyl and 5-FAM as the donor and acceptor pair with excitation 485nm and emission 530nm, making them extremely sensitive, and better than many of the available MCA/Dnp substrates.

PEPDAB008 is based on a substrate developed previously by researchers: Bickett DM, et al., A high throughput fluorogenic substrate for interstitial collagenase (MMP-1) and gelatinase (MMP-9) Anal Biochem. 1993 Jul;212(1):58-64. The BioZyme Inc. substrate is more sensitive than the one described by Bickett, et al, in that the fluorophore pair we use is Dabcyl/Fam. It is processed well by MMP9, MMP1 and other MMP enzymes. However,  the ADAM family of metalloproteinases do not process this substrate, thereby making it attractive for use with PrAMA. See publications 3-7.

PEPDAB011 or Flsub1 is extremely sensitive for MMP2, MMP9 and MMP13 and is highly selective over MMP1 or collagenase 1 and MMP3 or stromelysin 3. In addition, the substrate has 100 fold selectivity over other MMPs such as MMP8, MMP14, and MMP12.
PEPDAB052 is one of the most selective MMP9 substrate provided by BioZyme Inc. It is almost as sensitive as Pepdab011, but it has better selectivity against MMP2 and similar selectivities against many of the other MMP and ADAM family metalloproteinases. This MMP substrate was used in the proteolytic activity matrix analysis experiments (PrAMA). However, this fluorescence substrate can also be used individually to assess activity of MMP9.

PEPDAB016 is a very good MMP2 substrate (gelatinase A) and is more selective for this enzyme than MMP9 (gelatinase B). For specificity constants and use with biological samples. This MMP2 substrate was also used in the proteolytic activity matrix analysis experiments (PrAMA) described by Miller et al.,.

PEPDAB017 is a selective MMP9 substrate and has the opposite selectivity of PepDab016 in that it is selective over MMP2 for MMP9.

PEPDAB061 is a substrate sequence based on a selective biosensor for MT1-MMP or MMP14 and is a useful substrate to detect most MMP activities. We are currently offering a new promotional price of 1mg, 80 USD.
